Biopolymer of alginate nature with a predominance of L-guluronic acid

Highly viscous polysaccharide (250–350 kDa) of an alginate nature with a predominance of α-L-guluronic acid (M/G = 0.22) was obtained from Azotobacter vinelandi. The yield of polysaccharide was 20.5 ± 0.5 g/l when cultured in a medium containing molasses at a viscosity of the cultural liquid of over 30000 cSt. The biopolymer is stable at pH 4.0–9.0 in a wide temperature range and well soluble in highly mineralized water; it retains a high viscosity level and can be used in the petroleum industry for enhanced oil recovery.     

LncRNA NEAT1 promotes docetaxel resistance in prostate cancer by regulating ACSL4 via sponging miR-34a-5p and miR-204-5p

Docetaxel resistance remains one of the main problems in clinical treatment of metastatic prostate cancer (PCa). Previous studies identified differently expressed lncRNAs in docetaxel-resistant PCa cell lines, while the potential mechanisms were still unknown. In the present study, we found NEAT1 was expressed at high levels in docetaxel-resistant PCa clinical samples and related cell lines. When knockdown NEAT1, cell proliferation and invasion in docetaxel-resistant PCa cells in vitro and in vivo were downregulated. Our further researches explained that NEAT1 exerts oncogenic function in PCa by competitively ‘sponging’ both miR-34a-5p and miR-204-5p. Inhibition of miR-34a-5p or miR-204-5p expression mimics the docetaxel-resistant activity of NEAT1, whereas ectopic expression of miR-34a-5p or miR-204-5p attenuates the anti-drug function of NEAT1 in PCa cells. Besides, we also found ACSL4 is a target of both miR-34a-5p and miR-204-5p, and ACSL4 was also inhibited by miR-34a-5p and miR-204-5p. Moreover, suppression of miR-34a-5p or/and miR-204-5p greatly restrained the expression of ACSL4 upon NEAT1 overexpression. Joint expression of miR-34a-5p and miR-204a-5p synergistically decreased the expression of ASCL4, indicating miR-34a-5p and miR-204a-5p collaboratively inhibit the expression of ACSL4. Innovatively, we concluded that NEAT1 contributes to the docetaxel resistance by increasing ACSL4 via sponging miR-34a-5p and miR-204-5p in PCa cells.     

A NEW RHEOCRICOTOPUS THIENEMANN & HARNISCHIA (DIPTERA: CHIRONOMIDAE) FROM TAIWAN PROVINCE CHINA

Abstract  Rheocricotopus (Psilocricotopus) taiwanensis sp. n. from Taiwan, China, is described as male imagines. The species is allied to R. (P.) chalybeatus (Edwards) but it is easily separated from the latter in lacking setae in all reins, much lower AR and more pronounced crista dorsalis in gonostylus. The genus is new to Taiwan Province, China. The specimen is deposited at the Department of Biology, Nankai University, Tianjin.     

粘虫幼虫血淋巴中的凝集素

粘虫Mythlmna separata Walker幼虫血淋巴中含有凝集某些脊椎动物红细胞的凝集素,凝集活性可被乳糖、岩藻糖或神经氨酸抑制.用CNBr-sepharose 4B 进行亲和层析从血淋巴中分离的凝集素成分比较复杂,聚丙烯酰胺凝胶电泳显示三条区带,SDS聚丙烯酰胺凝胶电泳出现6个亚基,亚基分子量分别为71000、65000、56000、35000、33000及31000道尔顿.     

In situ hybridization to metaphase chromosomes in six species ofPhaseolus andVigna using ribosomal DNA as the probe

In situ hybridization with a biotin-labeled rice ribosomal DNA (rDNA) probe to the somatic metaphase chromosomes of six species ofPhaseolus andVigna (P. angularis, P. calcaratus, P. coccineus, P. vulgaris, V. sesquipedalis andV. sinensis) was done to determine the sites of rDNA. Hybridization signals were present in the terminal and subterminal chromosome regions of each of the six species. The number of rDNA sites was two inP. angularis andP. calcaratus, four inP. coccineus andP. vulgaris, and six inV. sesquipedalis andV. sinensis.     

几种氨基酸─铜（Ⅱ）配合物的超氧化物歧化酶活性

用四氮唑蓝光化学还原法对所合成的ＫＣｕ（ＩＤＡ）（Ｓｅｒ）·２Ｈ２Ｏ、ＫＣｕ（ＩＤＡ）（Ａｌａ）·Ｈ２Ｏ、Ｃｕ（ＩＤＡ）（ｅｎ）、ＫＣｕ（ＩＤＡ）（Ｇｌｙ）·Ｈ２Ｏ和Ｃｕ（ＩＤＡ）·２Ｈ２Ｏ（ＩＤＡ＝Ｎ（羧基甲基）－甘氨酸,Ｓｅｒ＝丝氨酸,Ａｌａ＝丙氨酸,ｅｎ＝乙二胺,Ｇｌｙ＝甘氨酸）等５种氨基酸─铜（Ⅱ）配合物进行了活性测定,发现它们均具有天然超氧化物歧化酶活性,其活性依次为０．３４、０．４５、０．５０、０．５４、０．７２Ｃｕμｍｏｌ·Ｌ－１。     

钙／钙调素依赖性蛋白激酶对17．7kD和6kD胰腺蛋白的磷酸化

本文报导了胰腺提取物中两种可被钙／钙调素依赖性蛋白激酶磷酸化的热稳定蛋白。ＳＤＳ－ＰＡＧＥ测定其表观分子量分别为１７．７ｋＤ和６ｋＤ。经钙／钙调素依赖性蛋白激酶磷酸化后,其最大磷酸参入量为８．８μｍｏｌ／ｇ蛋白。同时磷酸化作用导致１７．７ｋＤ蛋白在ＳＤＳ－ＰＡＧＥ中迁移率发生变化。本文还进一步分析了各种阳离子对磷酸化的影响,并对此两种蛋白可能具有的生理功能进行了初步探讨。     

SLE血清中抗-DNA抗体分离和性质的研究

本文用国产高分子树脂(T)接枝小牛胸腺DNA,通过亲合层析从系统性红斑狼疮SLE患者血清中纯化出抗-ds DNA抗体和抗-ss DNA抗体。酶联免疫吸附分析(ELISA)的研究表明:SLE抗-DNA抗体和DNA结合的差异性很大,是高度非均一性的。抗-ss DNA抗体不仅组成成分比抗-ds DNA抗体复杂,ss DNA/抗-ssDNA亲合能力也明显高于ds DNA/抗-ds DNA。纯化的抗-DNA抗体以IgG类抗体占主导,同时也有其它类型抗体存在(例如IgM等)。抗-ds DNA抗体有较抗-ss DNA抗体高的IgG含量(两者的IgG/IgM分别是7.0和4.0),说明IgG抗-DNA抗体更倾向于同dsDNA结合。
